Dynamic interconversion and phototransformation proces - ses of protochlorophyllide complexes during greening

چکیده
Photoreduction and interconversion processes of different protochlorophyllide complexes were studied in epicotyl segments of dark-germinated pea (Pisum sativum L.) seedlings illuminated with low-intensity white or 632.8 nm laser light. Analyses of 77 K fluorescence emission spectra showed the direct phototransformation of the monomer, 636 nm emitting complex. The 629 nm emitting complex containing also monomer pigment, regenerated all longer wavelength, flash-photoactive complexes. A dynamic transformation of the protochlorophyllide complexes was also detected during the illumination and/or the subsequent dark-incubation. On the basis of these results, the protochlorophyllide phototransformation schemes can be completed with new pathways. Acta Biol Szeged 49(1-2):219-220 (2005)

Red Light Induced Change in the Saponin Content of Prolamellar Bodies in A vena sativa
Etioplast, Protochlorophyllide, Prolamellar Body, Saponin, Red Light Exposure to red light of high intensities leads to saponin and prolamellar body degradation in etiolated Avena sativa seedlings. Exposure to red light of low intensities increases the saponin percentage found in prolamellar bodies without changing the overall saponin content. Proto chlorophyllide is assumed to act a decisive ...
متن کاملActivation volume and activation energy of the protochlorophyllide phototransformation and the Shibata shift determined with high pressure fluorescence spectroscopy
The kinetics of protochlorophyllide phototransformation and of the subsequent blue shift were studied in homogenates prepared from etiolated wheat leaves using fluorescence spectroscopy at high pressures. The pressure dependence of the processes was studied between 0.1 and 400 MPa. The activation volumes were calculated from the pressure dependence of the kinetical curves of the processes and w...
متن کاملDoes a light-harvesting protochlorophyllide a/b-binding protein complex exist?
Recent in vitro studies have led to speculation that a novel light-harvesting protochlorophyllide a/b-binding protein complex (LHPP) might exist in dark-grown angiosperms. Structurally, it has been suggested that LHPP consists of a 5:1 ratio of dark-stable ternary complexes of the light-dependent NADPH: protochlorophyllide oxidoreductases A and B containing nonphotoactive protochlorophyllide b ...
متن کاملChloroplast Biogenesis: XX. Accumulation of Porphyrin and Phorbin Pigments in Cucumber Cotyledons during Photoperiodic Greening.
A study of greening in cucumber (Cucumis sativus L.) cotyledons grown under a light (14-hour) dark (10-hour) photoperiodic regime was undertaken. The pools of protoporphyrin IX, Mg-protoporphyrin IX monoester, protochlorophyllide, and protochlorophyllide ester were determined spectrofluorometrically. Chlorophyll a and b were monitored spectrophotometrically. Pigments were extracted during the 3...
متن کاملFunctional analysis of isoforms of NADPH: protochlorophyllide oxidoreductase (POR), PORB and PORC, in Arabidopsis thaliana.
NADPH:protochlorophyllide oxidoreductase (POR) catalyzes the light-dependent reduction of protochlorophyllide. To elucidate the physiological function of three differentially regulated POR isoforms (PORA, PORB and PORC) in Arabidopsis thaliana, we isolated T-DNA tagged null mutants of porB and porC. The mature seedlings of the mutants had normal photosynthetic competencies, showing that PORB an...
